Characterization of Cellulase from Paenibacillus polymyxa PC-1 Isolated from Soil

Abstract
Using LB-Agar plate containing carboxymethyl-cellulose (CM-cellulose) and trypan blue, we screened six cellulolytic microorganisms from soil at Sangju-si, Gyeongbuk, Korea. The isolated microorganisms were identified with four kinds of Bacillus subtilis, one of Bacillus amyloliquefaciens, and one of Paenibacillus polymyxa by the analysis of 16S rRNA gene. Among them, one strain showing the highest enzyme activity for CM-cellulose was selected and was named as a Paenibacillus polymyxa PC-1. The cellulase activity of the strain showed the maximum activity at pH 6.0, 50¡ÆC, and 1.5% CM-cellulose. In the investigation of pH and temperature stability, the cellulase was stabled at pH 5.0~7.0 and 50¡ÆC for 30 min perfectly. It exhibited the highest substrate specificity against CM-cellulose. And, the enzyme activities for Alkali swollen cellulose, Sigmacell-cellulose, Alpha-cellulose and Avicel were approximately 11%, 8.1%, 6.8% and 5% of activity for CM-cellulose, respectively. Under the conditions of pH 6.0 and 50¡ÆC, the cellulase activity was saturated after 30 min of reaction with 1.5% CM-cellulose.
